چکیده

The maltose α-amylase AmyM from Bacillus stearothermophilus can be used for flour modification, baked goods preservation, and production. Here, we optimized the recombinant expression of in subtilis WB800 via several strategies. By screening optimal promoter, a double promoter combination (P43 PamyL) could improve level by 61.25%, compared with strong P43. Then, secretion efficiency over-expressing molecular chaperone prsA gene. SDS-PAGE results suggested that over-expression to extracellular environment. enzyme activity was increased 101.58% control strain. To further AmyM, introduced hemoglobin gene Vitreoscilla (vgb) into revealed introduction vgb promote transcription translation B. subtilis. This may due increasing intracellular NADPH NADP+ caused vgb. this strategy, 204.08%. Finally, found showed an temperature 65 °C pH 5.5. Our present provided effective strategy heterologous

Bottleneck in secretion of α-amylase in Bacillus subtilis

Amylase plays an important role in biotechnology industries, and Gram-positive bacterium Bacillus subtilis is a major host to produce heterogeneous α-amylases. However, the secretion stress limits the high yield of α-amylase in B. subtilis although huge efforts have been made to address this secretion bottleneck. In this question-oriented review, every effort is made to answer the following que...

Production, purifi cation, and characterization of α-amylase by Bacillus subtilis and its mutant derivates

Th e eff ects of various carbon and nitrogen sources on production of α-amylase by Bacillus subtilis and its mutant derivates were investigated. Th e maximum production of α-amylase by all strains was obtained in the presence of mesoinositol as the carbon source. Th ere was no more signifi cant increase in enzyme yield in the case of the supplementation of nitrogen sources, whereas malt extract...

Enzymatic synthesis of glycosylated puerarin using maltogenic amylase from Bacillus stearothermophilus expressed in Bacillus subtilis.

BACKGROUND The maltogenic amylase from Bacillus stearothermophilus (BSMA) is a valuable biocatalyst that has been used to transglycosylate natural glycosides to improve solubility. To ensure safety, BSMA was produced in Bacillus subtilis, using new shuttle vector-based expression vectors. The transglycosylation of puerarin was also conducted with crude BSMA and analyzed. RESULTS Two expressio...

Heterologous expression, biochemical characterization, and overproduction of alkaline α-amylase from Bacillus alcalophilus in Bacillus subtilis

BACKGROUND Alkaline α-amylases have potential applications for hydrolyzing starch under high pH conditions in the starch and textile industries and as ingredients in detergents for automatic dishwashers and laundries. While the alkaline α-amylase gains increased industrial interest, the yield of alkaline α-amylases from wild-type microbes is low, and the combination of genetic engineering and p...

Role of maltogenic amylase and pullulanase in maltodextrin and glycogen metabolism of Bacillus subtilis 168.

The physiological functions of two amylolytic enzymes, a maltogenic amylase (MAase) encoded by yvdF and a debranching enzyme (pullulanase) encoded by amyX, in the carbohydrate metabolism of Bacillus subtilis 168 were investigated using yvdF, amyX, and yvdF amyX mutant strains. An immunolocalization study revealed that YvdF was distributed on both sides of the cytoplasmic membrane and in the per...
